Анализ сетей коэкспрессии генов и белок–белковых взаимодействий с участием натрий-зависимого фосфатного транспортера NaPi2b в нормальных и опухолевых тканях яичника

Аннотация
Исследование посвящено сравнительному анализу сетей коэкспрессии гена SLC34A2 натрий-зависимого фосфатного транспортера NaPi2b с генами гликозил- и сульфотрансфераз и взаимодействия продуктов этих генов в нормальных и опухолевых клетках яичника для выявления белков, ответственных за аберрантное гликозилирование транспортера NaPi2b при злокачественной трансформации клеток. Данные по экспрессии 214 генов гликозил- и сульфотрансфераз и гена SLC34A2 в нормальных и опухолевых клетках были получены из исследования E-MTAB-3732 (ArrayExpress, n = 579), а данные о 7025 белок–белковых взаимодействиях из открытых баз данных. Построенные на основе собранных данных сети показали различия в паттернах коэкспрессии генов и взаимодействия их продуктов. Коэффициент кофенетической корреляции сетей в нормальных и опухолевых клетках яичника составил 0.17. Ключевые гены B3GNT2, GCNT1, ST3GAL1, ST6GAL1 входят в состав крупнейшего модуля вместе с геном SLC34A2 в сети коэкспрессии генов и белок–белковых взаимодействий в опухолях. Сделано предположение, что профили экспрессии и взаимодействий продуктов генов ST6GAL1, B3GNT2, GCNT1, ST3GAL1 могут быть связаны с аномальным гликозилированием NaPi2b в опухолевых клетках. Дальнейшие исследования этих белков важны для понимания роли гликозилирования в изменениях конформации большого внеклеточного домена транспортера NaPi2b и разработки противоопухолевых препаратов против потенциального опухоль-специфического эпитопа МХ35 транспортера NaPi2b.
